  • Repair Skin Barrier

Plamed has done the test of  in vitro 3D epidermis skin model repairing efficacy, using the anionic surfactant Sodium lauryl sulfate (SLS) to stimulate the 3D epidermal skin model to construct an in vitro skin injury model. After PMRepair® Centella sample is applied, the tissue morphology, filaggrin (FLG), loricrin (LOR) and transglutaminase 1 (TGM1) are used to evaluate the repairing efficacy of PMRepair® Centella.

The test results are as follows:

 

Based on the 3D epidermal skin model (EpiKutis®), compared with the control group, the tissue morphology of the sample PMRepair® Centella at a concentration of 0.15% was significantly improved, and the thickness of the living cell layer was significantly increased, with an increase rate of 49.39%. Filaggrin (FLG), loricrin (LOR), and transglutaminase 1 (TGM1) contents all increased significantly, with the increase rates being 150.00%, 646.15%, and 537.14% respectively. It indicates that PMRepair® Centella can improvetissue morphology at 0.15% concentration, promote skin tissue recovery and increase the content of barrier-related proteins (FLG, LOR, TGM1) to achieve repairing effects.

Based on skin barrier permeability test of zebrafish embryo, PMRepair® Centella has a significant skin barrier repair effect at a lower dose of 0.1%, the repairing rate is 53%; while Asiaticoside (95%) has no repairing effect or has a weak repairing effect at either low or high doses.

  • Anti-inflammation & Anti-bacteria

NF-κB signaling pathway is closely related to inflammatory response and plays an important role in regulating cellular inflammatory response: when cells are stimulated, NF-κB can be activated, thereby causing inflammatory factors TNF-α, IL-1β, IL-2, IL-6, nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2)’s over-expression.

Relevant studies have found that Centella asiatica extract can down-regulate the expression of nitric oxide (NO), TNF-α, and IL-6 by inhibiting the NF-κB signaling pathway, thereby exerting anti-inflammatory effect. Centella asiatica extract can also play an anti-inflammatory role by regulating the heme oxygenase 1 (HO-1) signaling pathway, inhibiting the production of pro-inflammatory factors and the activity of peroxidase.

In addition, Centella asiatica extract has a certain inhibitory effect on Pseudomonas aeruginosa, Staphylococcus aureus, and acne bacillus. Asiatic acid can dissolve the outer wax film of bacteria to be killed by other drugs or body defense tissues. Asiatic acid and glycosides for external use can promote wound healing, stimulate granulation, promote epidermal keratinization or epidermal regeneration, and help generate connective tissue. Centella asiatica extract can reduce the production of pro-inflammatory mediators (IL-1, MMP-1), improve and repair the skin’s own barrier function, thereby preventing and correcting skin immune dysfunction.

  • Anti-oxidation

Centella asiatica extract also has a strong antioxidant effect, which can inhibit free radical activity. Oxidation is one of the important causes of skin aging. Radiation and air pollution in the external environment will cause the body to produce free radicals. Free radicals are active substances that lack electrons. After entering the human body, they easily snatch electrons from cells and cause cell distortion. This is the root cause of human aging and disease. Specialists studied the antioxidant activity and total phenolic compounds of the roots, leaves and petioles of four species of Centella asiatica, and the results showed that the leaves and roots of Centella asiatica had high antioxidant activity, and their antioxidant activity was closed to that of Tocopherol.

Note: PMRepair® Centella at a concentration of 0.15% has been tested with very good efficacy results based on 3D epidermal skin modeling, so the recommended dosage is 0.1%-0.2%.

The water solution of PMRepair® Centella is clear and transparent, but the solution will be cloudy and opaque when stand for a long time.

Therefore, the following dissolution methods can be chosen:

 35% butylene glycol, 65% deionized water. The solution is clarified and transparent after a long time.

 Pure water. At 0.1% and 0.2% concentration and PH value below 6, the aqueous solution is clear and transparent.
